To “What’s your brother like?”, the answer could be: He is like a chick. (mentally)
To “What does your brother look like?”, the answer has to be made with respect to the appearance: e.g., He looks like a man but is actually a chick.

“What does your brother look like” - is question about appearance. It is possible with “look” only. In the answer you can use " He is …" or “He looks like …”
“What’s your brother like” == What is your brother like". The answer would be “He is like …”
