To “What’s your brother like?”, the answer could be: He is like a chick. (mentally)
To “What does your brother look like?”, the answer has to be made with respect to the appearance: e.g., He looks like a man but is actually a chick.

“What’s your brother like?” is more about personality.

“What does your brother look like?” is about appearances.

“What does your brother like” has absolutely different meaning. The answer would be “My brother likes apples”.

“What does your brother look like” - is question about appearance. It is possible with “look” only. In the answer you can use " He is …" or “He looks like …”

“What’s your brother like” == What is your brother like". The answer would be “He is like …”
