The good speakers must possess sincerity, good platform manners/a good platform manners, and the ability to express his thought.
Which one is correct?

I don’t understand the meaning of ‘good platform manners’ at all (though it definitely would not take an article). ‘Good speakers’ being plural, would not take an article either, but ‘the good speaker’ would be okay. ‘Thought’ should be ‘thoughts’.

The article applies to the head noun in the phrase, which is “manners”, and “a … manners” is not possible. “a good platform manner” would be possible.

“thought” at the end should usually be “thoughts”.

“The good speakers … ability to express his thoughts” does not work. You can say:

“Good speakers … ability to express their thoughts” (good speakers in general; this is the most usual combination, and probably what you mean).
“The good speakers … ability to express their thoughts” (some specific good speakers).
“A good speaker … ability to express his thoughts” (any good speaker).
“The good speaker … ability to express his thoughts” (any good speaker).

Some people would quible about the use of gender-specific “his”, but that debate is very well covered elsewhere.

I understood it to mean good manners when on the platform (= stage, podium) speaking.

Thinking about this some more, I think what you probably mean is “a good platform manner”.

“A good speaker must possess sincerity, a good platform manner, and the ability to express his/her thoughts.”
Is this sentence OK?

“his or her” is preferred in polished writing.

As has been mentioned, “thought” should be “thoughts”, and you probably mean “Good speakers must…” rather than “The good speakers must…”.

“sincerities” is wrong. “abilities” is not natural in this context.

There are a few dozen Google hits for “a good platform manner” in the relevant sense. My opinion is still that it is an acceptable phrase.
