As you explained that on leave means on vacation. So can i say " I want to spend my summer to be on leave" Or I’m not stay at home, i 'm on leave?

You could say spend the summer on leave. Your second sentence: I’m not staying at home, I’m on leave doesn’t really make sense because the two states are not really related. You could say: I’m not staying at home because I’m going on holiday.
