Do these two sentences have exactly the same meaning?

In normal conversation they have the same meaning. Although “my ex-boss” is more common.

You asked if they have exactly the same meaning. The first sentence slightly implies that you have more than one ex-boss. In practice, nobody will think about it that hard. Both sentences will be interpreted the same.
