Driver: No, then you have to transfer onto another bus.
Man: You mean I have to take another bus? I have to transfer again.
Driver: Yes.
Man: Oh, I think that’s going to take me a little too long. I’m going to take my car today. Thanks.

What does " take me a little too long" mean?

Does it mean “take a little too long of my time”?

Like this: I think that’s going to take a little too long of my time.

The phrase “take me a little too long” means that the trip will require more time than the speaker finds acceptable.

Your version “take a little too long of my time” is not quite natural. A better phrasing would be:
:small_blue_diamond: “I think that’s going to take a little too much of my time.”

However, in the original sentence, “take me a little too long” is already idiomatic and commonly used in spoken English.
