If the question is in the present simple and involves a verb other than “to be,” it starts with “Do” or “Does.” Use “Does” if the subject is third-person singular (he, she, it, John, Sally, the cat, etc) Use “Do” everywhere else.

Examples:

Do you like tennis?
Does John work with you?
Do they live next door?

Are you American?
Is John hungry?
Are they awake?
